Fluorescent tags are major tools in biological and biomedical research. We propose a unique structural modification applicable to the most common organic fluorophores, in order to generate analogues endowed with the new properties of photoactivability, covalent capture and fluorogenesis (PCCF). These
tags, after conjugation to small molecules or macromolecules of interest (medicine, natural protein product, RNA), would make it possible to identify their targets and biological partners. This approach relies on interactomic and imaging studies, with an emphasis on the super-resolution microscopy expected from these PCCF tags. The objectives of this project will first be the synthesis of a portfolio of PCCF tags and then their functional screening on simple biological models. A selection of optimal tags will then be validated in reference paradigms involving known ligand-partner couples, then used to
explore the interactome of new relevant ligands.
